GW9662 blocked DHA- and EPA-induced increases in cellular and secreted adiponectin by inhibiting PPARγ. 3T3-L1 adipocytes were incubated with 100 μmol/L of DHA (or EPA) in the presence or absence of 10 μmol/L of GW9662, or GW9662 alone for 24 h. BSA treatment served as the control. Cellular PPARγ (a, c, d), adiponectin (b, c, e) and secreted adiponectin (f) were assessed. Cellular PPARγ and adiponectin in treatment groups were normalized to the control with β-actin worked as the internal reference.
